Abstract This work aims to study the dormancy and the rest period in certain varieties of gladiolus corms in relation to some physiological activities that occur during that state in different seasons. The respiration of the corms is measured as an indicator of the metabolic activities during this period. It is measured at two-weeks intervals beginning after the end of flowering until the-end of the rest stage, in both winter and summer seasons. The measurements of the respiration rates are carried out by the flow stream method. The tetrazolium is also used to indicate certain enzymatic activities during the dormancy period. It is conducted by using pieces of two grams of the corms with 10 ml of 1% water solution 2, 3, 5- triphenyltetrazolium chloride (TTC) for a period of two hours at 40 C. The residue is extracted by acetone and dissolved in butylalcohol. The quantity of the residue in the pieces is determined calorimetrically by the aid of a standard curve for known weights of the residue. |
