Abstract This work was carried out on the submandibular gland of adult male albino rat, 25 animals were used, 5 were used as control and the remaining animals were castrated and divided into 5 groups, the first two groups were killed one week and two weeks after castration, the remaining operated groups were treated with testosterone hormone by dose, 4 mg/kg body weight every other day and their submandibular glands were removed after one week from treatment in the third group, two weeks in the fourth group and four weeks in the fifth group. Each submandibular glands were divided into two parts, one part was fixed in acetone for alkaline phosphatase enzyme activity, the other port was fixed in bouin solution and used to examine the following stains. 1. Hematoxylin and eosin stain. 2. Mallory stain. 3. Periodic acid schiff’s reaction. 4. Alcian blue of Ph 2.5.
