Abstract

Fibrates, which are hypolipidaemic drugs known as synthetic PPAR-α ligands, have been shown to offer protection against I/R injury. The present study was undertaken to study the effect of PPAR-α agonists, bezafibrate and clofibrate in acute and sub-acute prophylactic treatment against gastric ulcer model in rats. Animals were divided into 12 groups : animals received saline as a sham operation group (group 1), a I/R group received saline (group 2), a I/R group received corn oil (group 3), animals treated with bezafibrate ( 50mg/Kg) before I/R (group 4), animals treated with clofibrate ( 50mg/Kg) before I/R (group 5), animals treated with ranitidine (75mg/kg) before I/R (group 6), animals received saline single dose /day for three days as a sham group (group 7), animals received saline single dose /day for three days before I/R (group 8), animals received corn oil single dose /day for three days before I/R (group 9), animals treated with bezafibrate ( 50mg/Kg/day) for three days before I/R (group 10), animals treated with clofibrate ( 50mg/Kg/day) for three days before I/R (group 11), animals treated with ranitidine (75mg/kg/day) for three days before I/R (group 12).Acute and sub acute effect of Bezafibrate, clofibrate and ranitidine decreased ulcer score , ulcer index, titratable acidity, peptic activity. glutathione-S-transferase and lipid peroxides contents, while systolic, and diastolic blood velocity and SOD activity were increased significantly. The histopathological results showed that, Ischemia/reperfusion showed extensive massive necrosis , ulceration, severe edema and inflammatory infiltration. Acute effect of Bezafibrate, clofibrate and ranitidine caused moderate protection of the stomach against I/R induced tissue necrosis, ulceration and inflammatory infiltration, while sub acute effect produced marked protection of the gastric mucosa. Our data indicate that Bezafibrate, clofibrate and ranitidine produced significant gastroprotective effect against I/R-induced gastric ulcer
