Abstract This work was undertaken to study the histological, histochemical and ultra structural changes in the rats liver after injection of cisplatin alone and cisplatin associated with vitamin E. Thirty adult male albino rats were used in this study, they were classified into three groups.Small liver slices were prepared and stained with Haematyoxylin & eosin and periodic acid Schiff?s stains. Fresh frozen cryocut sections (1012 micron) were prepared and treated for demonstration of acid phosphatase and succinic dehydrogenase enzyme activities in the liver. Small fragments of the liver were processed to obtain ultra thin sections which were stained and examined with transmission electron microscope. Haematoxylin and Eosin (H&E) sections of cisplatin treated animals showed that the liver lesions were progressive and dose dependant in the form of inflammatory cell infiltration, congestion in the central viens & blood sinusoids and apoptosis. Periodic acid schiff`s stained sections of cisplatin treated rats livers showed gradual and progressive decrease in glycogen granules. The histochemical study showed gradual decrease in S.D. activity accompanied with gradual increase in the acid phosphatase reaction in the hepatocytes all over the hepatic lobules Electron microscopic study of the livers of rats administrated cisplatin exhibited marked decrease in the glycogen granules and mitochondrial changes which included decrease in the number, deformity in shape, disruption of the membrane and loss of the cristae. Partial loss of the microvilli at the sinusoidal surface was observed. In addition, increase in the number of lysosomes was also seen.These changes were mild in the hepatocytes of rats treated with vitamin E before administration of cisplatin |
