Abstract Twenty aault alblno rats were used in this study Paraffin sections were prepared from the diaphragm of these animals, stained lith haematoxylin and eosin and phosphotungstic acid haematoxylin. In these sections the skeletal muscle fibers appeared long cylinderical multinucleated cells. The muscle nuclei were flattenned and peripheral in position under the sarcolemma. Fibroblasts and satellite cells appeared between the muscle fibers. Transverse sections showed the polygonal ou~line of the majority of the muscle fibers of the diaphragm, small sized muscle fibers with heavily clustered myofibrils and large sized muscle fibers with finely dispersed myofibi’ils appeared in the section. Cross striations of alternating dark and light bands in the muscle fibers were clear in phosphotungstic acid haematoxylin stained preparations. Frozen sections were prepared, used to study the activity of succinic dehydrogenase lSDHase), acetylcholinesterase lkCh-E) and non-specific esterase (H-SE Succinic dehydrogenase activity revealed three types of muscle fibers in the diaphragm of the rat. Small sized muscle fibers lType Cj with strong positive activity and these I muscle fibers corresponded to the slow oxidative type, large sized muscle fibers lType A), with a weak activity and these corresponded to the fast glycolytic type and intermediate muscle fibers (Type B) with a moderately positive activity . |
