Abstract This work studied the neurodegeneration and demyelination in the adult hippocampus, corpus callosum, and optic nerve in the cuprizone rat model of multiple sclerosis and the possible protective effect of thymoquinone versus platelet rich plasma derived lyophilized growth factors. A total number of 40 adult (200-250 gm) male albino rats were used in this study and divided equally into 4 equal groups; a control group, they recieved nothing. The CPZ group recieved cuprizone in a dose of 0.6 g added to 100 g of chow (0.6%) daily for 4 weeks. The CPZ+TQ group which received (0.6%) cuprizone on concomitant with thymoquinone (20 mg/kg b.w. dissolved in corn oil (0.5 ml/rat) by gastric tube) daily for 4 weeks. The CPZ+ PRP d. LGF which received (0.6%) cuprizone on concomitant with lyophilized growth factors (LGF) (250 μL dissolved in distilled water, intraperitoneally) twice/ week for 4 weeks. At the end of the experiment, the samples of the brain and optic nerves were extracted from the skull and proceeded for light and electron microscopic study and immunohistochemical examination of the hippocampus, corpus callosum and optic nerve to study their structure and detect any changes in the different examined groups.
