Abstract Introduction Prostate cancer is the second most common malignancy in men and the fifth leading cause of death from cancer worldwide. In Egypt, prostate cancer ranks fifth among the most common types of cancer among males. Recent studies indicate the involvement of genetic factors in increasing the risk of prostate cancer. Aim of the study The present study aimed to evaluate the potential association between 8q24 (rs1447295) and PON1 (L55M) single nucleotide polymorphism (SNP) and prostate cancer risk in a cohort of Egyptian male prostate cancer patients. Patients and methods This study was conducted on a group of 102 Egyptian males, including 51 patients with prostate cancer from Alexandria Kidney and Urinary Tract Hospital and 51 other males with benign prostatic hyperplasia after it was confirmed that they didn’t have prostate cancer. DNA was extracted using the DNA salting out technique, then all DNA samples were analyzed using PCR-RFLP technique and using appropriate cutting enzymes. Results Statistical comparisons between the prostate cancer patients in the current study and the control sample of non-afflicted patients showed that there was a statistically significant difference, indicating an association of the A allele in the 8q24 gene rs1447295 with the risk of prostate cancer (P=0.0001) and an association of the M allele in the PON1 L55M gene.
