Abstract

Poorly soluble medications have traditionally difficulty being administered through the eyes. Drugs are unable to remain within the cornea and precorneal areas of the eye cavity due to the different anatomical and physiological barriers there. In this work, the Nano system was able to administer the insufficiently soluble Acyclovir via the eye. Our Nano system is made of PVP\HA with pyridoxine. The stability of Acyclovir in a new polymeric hydrogel ophthalmic formulation towards gamma radiation sterilization was studied using UHPLC method. The gamma radio-sterilization of the formulation may result in degradation of the main active material (Acyclovir). So, the free radicle scavenger Pyridoxine was added to protect the drug from degradation during sterilization. The advantages of using these new drug forms of controlled release are, among others, increasing bioavailability of substance through extending the time of its contact with cornea—which can be achieved by effective adhesion to the corneal surface, the possibility of targeted therapy preventing the loss of drug to other tissues, ensuring patient’s comfort when applying the drug form and during the whole therapy, and increasing resistance to eye defense mechanisms, like tearing. PVP solutions in water were exposed to radiation to create these hydrogels. Macro radicals are formed when PVP interacts with the hydroxyl radicals released during water radiolysis. Therefore, it might be said that using radiation to create medication delivery systems is a manageable method.
