Abstract Various approaches of pulp regeneration technique can differently effect on pulpal healing mechanism. The present study evaluated the effect of photobiomodulation, and exosomes derived from dental pulp stem cell of the dogs on direct pulp capping through biochemical analysis and gene expression . Methods: Twenty-four healthy mature mongrel dogs were divided into eight groups ;group I, control; group II, photobiomodulation ; group III, tricalcium silicate cement ; group IV, stem cells ; group V, tricalcium silicate cement+PBM ; group VI, stem cells +PBM ; group VII , stem cells + tricalcium silicate cement and group VIII, combination of stem cells + tricalcium silicate cement +PBM. Blood samples were collected from dogs at days 7 postoperatively for biochemicals and gene expression analysis. Biochemicals as total and ionized calcium, phosphorus, and alkaline phosphatase levels and the gene expression levels of matrix metallopeptidase 9, osteocalcin, transforming growth factor beta genes were measured .Also histological examinations was done for inflammatory response and dentine bridge formation. Results: Statistical analysis showed that there were significance differences between all groups in different assessed biochemicals and genes expression’s levels. The combination group of PBM+ stem cells + tricalcium silicate ( group VIII) showed the highest level of biochemicals and gene expression. Also, regarding the histological examination, group VIII showed the least pulpal tissue inflammation and highest dentine bridge thickness. Conclusions: PBM combined with stem cells and tricalcium silicate cement enhanced the mineral content like total and ionized calcium , phosphorus level and ALP as well as increased gene expression of MMP9, TGF- β, and OCN during pulp regeneration procedure in dogs, as well as less inflammatory pulp response and highest dentin bridge thickness. |
