Abstract SUMMARY This study was conducted to investigate the effect of extra virgin olive oil, trastuzumab and pegylated liposomal doxorubicin on liver, and kidney, lipid profile, glucose, hematological picture and histopathological changes, also the protective effect of extra virgin olive oil in combined therapy with trastuzumab and pegylated liposomal doxorubicin in mammary carcinoma. Sample were taken at day 8, 16 and 24. This study consists of 180 female Sprague Dawley-Rag2 rats weighting 130- 150-gram albino. The rats were randomly divided into main 8 groups: group (1): 20 female rats were administrated saline only 0.2ml daily for 30 days. group (2): 40 female rats 20female were administered (DAMBA) 60 mg/kg body weight in 0.2ml of sesame oil single dose intragastric for induction of mammary carcinoma. And 20female each rat was implanted subcutaneously BT-474 in the mammary fat pad of rat. group (3): 20 female rats were administered DAMBA) 60 mg/kg body weight in 0.2ml of sesame oil single dose intragastric for induction of mammary carcinoma as group 2 then followed by pegylated liposomal doxorubicin 30mg/m2 intravenously in tail vein at day1 and day14. group (4): 20 female rats were administered DAMBA) for induction of mammary carcinoma as group 2 then followed by pegylated liposomal doxorubicin 50mg/m2 intravenously in tail vein at day1 and day20. group (5): 20 female rats were implanted subcutaneously BT-474 in the mammary fat pad for induction of mammary carcinoma as group 2 then followed by intravenous injection of pegylated liposomal doxorubicin 30 mg/m2in tail vein at day 1 and day 14 and trastuzumab 4mg/kg intravenous for first dose thereafter 2mg/kg every week. |
