Abstract

Abstract
Background Prohibitin (PHB) is a protein that present in humans is encoded by the PHB gene. Prohibitin family is composed of two members, PHB1 and PHB2. PHB1 was described as a tumor suppressor gene for its ability to inhibit cell proliferation. Prohibitins (PHB1 and PHB2) have been proposed to play an important roles in cancer development and progression,PHB1 and PHB2 were found to be over expressed in a panel of leukemia and lymphoma cell lines compared to normal naïve peripheral blood mononuclear cells (PBMCs).
Aim The aim was to measuring the serum level of prohibitin in newly diagnosed multiple myeloma patient and correlating its level with prognostic markers and with patient outcome.
Patients and methods serum prohibtin level was measured by using enzyme-linked immunosorbent assay(ELISA) in 70 newly diagnosed multiple myeloma patients ,patients were chosen for the study ,more than or equal 18 years All patients were recruited from Clinical Hematology Department, Ain Shams University Hospital (Cairo, Egypt) over the period from April 2020 to April 2021 and followed for 1 year.
Results measurement of serum level of prohibitin in newly diagnosed multiple myeloma patients in study group which increase expression of prohibtin in multiple myeloma patient .There was significant correlation when compairing the serum prohibtin level with Age (years) it was [r=0.441; p-value <0.001]; S.creat (mg/dl) it was [r=0.643; p-value <0.001]; T.Ca (mg/dl) it was [r=0.432; p-value <0.001]; B2 microglobulin (ug/m) it was [r=0.873; p-value <0.001]; Plasma cells% in B.M it was [r=0.852; p-value <0.001]. This indicates the existence of a strong direct relationship, by increasing these previous factors, it leads to the increase of serum prohibitin level (ng/L).that there was significant correlation when compairing the serum prohibtin level with Hb. (g/dl), Plt (x10^3).which increase prohibtin leading to deacrease Hb and plt.here was significant correlation,but when compairing the serum prohibtin level with bence jones protein in the study group found increase prohitin in study group leading to increse bence jones protein in study group with (p-value p<0.05).
The study showed that increase serum level of prohibitin in newly diagnosed multiple myeloma patients have prognostic value which increase prohibtin level has bad prognosis in multiple myeloma patients.
Conclusion prohibitin has prognostic significance in newly diagnosed patient with multiple myeloma which increase prohibitin level leading to bad prognosis.
