Abstract Snake venom is a fascinating natural blend of proteins and peptides that is rich in variety and therapeutic potential. Much emphasis has been dedicated to the research of venom detoxification in various methods, as well as its biological significance. The aim of this study was to evaluate the anticancer potential of crude CV, irradiated Cerastes cerastes snake venom IRRV and propolis ethanolic extract &PEE related biological alterations. The study showed that for IRRV, there is a higher toxicity than CV for A549 and PC3 cell lines but CV-PEE showed a higher toxicity than IRRV and CV. The toxicity to A549 and PC3 cells was concentration and cell type dependent, in comparison to controls. The cell cycle analysis were monitored using flow cytometry and mRNA expression levels of the apoptosis-regulatory genes P53, Casp-3, and BCL-2 were determined by real-time PCR after treatment with IRRV, CV, PEE, and CV-PEE . Apoptotic genes showed a significant up-regulation of P53 and Casp-3 genes while the anti-apoptotic gene (Bcl-2) showed a down-regulation. IRRV showed the highest increase in P543 and Casp-3 and the lowest decrease in Bcl-2 for both PC3 & A549. Also, induced elevated DNA accumulation in the S phase post- PC3 cell treatment with IRRV and CV, as well as a significant DNA accumulation at G2/M phase after IRRV treatment of A549 cells. In contrast, PC3 cells showed a negligible cellular DNA accumulation after PEE treatment. Additionally, the level of oxidative stress can be measured by assessing the Glutathione reductase GR, Malondialdehyde MDA and Reactive oxygen species ROS levels. GR was reduced in case of PC3 and A549 cell treated with IRRV, CV, and PEE compared with its values in untreated cell control. The MDA values in both cells recorded a significant elevation post IRRV treatment compared to the rest of the treatment regimen and untreated cell control. Similarly, IRRV and CV-PEE showed obviously higher ROS values than PC3 and A549 cell treatments with CV and PEE. The conclusion, natural products as propolis may be an alternative way for detoxification of snake venom |
