The study deals with examined cattle in Al Baharia ,Al Kharga Oases and Siwa (Egypt) which were naturally infested with H. a. anatolicum. Preparation of different antigens from (eggs, larvae, nymphae and whole adult males, whole adult females, male salivary glands, female salivary glands (both fed for 5 days) and male midgut , female midgut as well as purified midgut peaks by gel filtration chromatography for immunization of rabbits in three experiments against tick infestation. The midgut from 2nd experiment and peak III of purified midgut were a significant reduction in the biological parameters of tick and effect in feeding and reproduction performance. Serum globulins were a significant increase in rabbits immunized with midgut in 2nd experiment and peak II then peak III of purified midgut in third experiment. Antibody levels were detected by ELISA with a significant increase in rabbits with midgut and peak III. Detection of polypeptide bands by using SDS-PAGE. By using western blot, the midgut antigens could protect rabbits from subsequent tick feeding and in considered the best antigen in the antigenic solutions in these study. Thus, a 59, 51, 33 and 20KDa (peakII &peakIII) might be diagnostic specific in midgut of H. a. anatolicum ticks while the polypeptide band with molecular weight 66 KDa was the most immunogenic epitopes might be play role in immunization and protection against H. a. anatolicum. Histological and TEM examination of salivary gland and midgut of female H. a. anatolicum 15 days fed on rabbits immunized with midgut antigen and control one.
