Abstract The aim of the present study was to compare the diagnostic accuracy of two indicators (PHH3 and AgNOR) in different grades of MEC and to correlate the sensitivity of their proliferative index with the histopathologic grades. A total 22 cases of MEC of salivary glands of different grades was used in this study. The collected cases were serially cut into 4 om thick sections and stained with (H&E) and were graded according to AFIP grading system Ten cases of H&E section of MEC were diagnosed as low grade tumors and ten cases of H&E section of MEC were diagnosed as High-grade . Paraffin blocks were cut into 4 micron thick sections and placed on positively charged slides for immunostaining procedures and prepared for PHH3 and AgNOR stain. The positive immunoreaction for PHH3 and AgNOR stain was assessed by transmission light microscope and the image analyzer computer system .The nuclear count of PHH3 immunostaining and AgNOR was calculated for five fields in each slide. Mean values of nuclear count were obtained for each group for statistical evaluation |
