Abstract

Polycystic ovary syndrome (PCOS) is a syndrome of ovarian dysfunction associated with oxidative stress. This study aimed to demonstrate the therapeutic role of selenium nanoparticles (SeNp) in PCOS and compare it with oral metformin treatment in experimentally-induced polycystic ovary in rats. Sixty adult female albino rats were divided as follows, twenty rats in group I (normal control), that were subdivided into subgroup Ia, sacrificed on day zero, and subgroup Ib, sacrificed at the end of the expirement. Forty rats, that received PCOS induction by testosterone ethanoate, were subdivided into 4 groups, each containing 10 rats, group IIa (PCOS) that received no treatment and were sacrificied right after PCOS induction, group IIb (recovery) that were sacrificied at the end of expirement, group III (metformin) received oral metformin for 21 days and group IV (SeNp) that received SeNp intraperitoneal injection for 21 days. Ovarian specimens were processed and stained with H & E, Periodic acid Schiff (PAS) stain, immunohistochemichal stains using TNF-alpha and IL-1 antibodies, also tissue biochemical study, hormonal blood tests, histomorphometric and statistical analyses were done.Metformin and SeNp improved histological architecture of the affected ovaries, restored normal follicular development and regular ovulation. Metformin and SeNp also regulated the disturbed hormonal blood levels and BGL, with significant reduction of random BGL in metformin treated group versus PCOS, recovery and selenium groups. SeNp enhanced the oxidative state enzymes, where it significantly reduced the elevated GPx and SOD tissue levels compared to metformin group, and decreased tissue inflammatory mediators TNF-alpha and IL-1 in the ovarian tissue examined
