Abstract The current study aims to investigate the effect of melatonin on the testis in rats exposed to gamma radiation. Male Wister rats were divided into 5 groups (n = 10) as follows: control group, sham group received melatonin solvent, two melatonin groups received melatonin for 3 and 14 days, two irradiated groups were exposed to whole-body gamma irradiation and assessed after 3 and 14 One day, two treatment groups received radiation and melatonin. Testicular injury in mice was masked by irradiation with a single dose of 4 gamma rays. The irradiation was performed with cesium-137 at a dose rate of 0.695 Gy/sec. Melatonin was administered orally at a dose of 25 mg/kg 30 min before irradiation. After 3 and 14 days after the combined treatment, mice were euthanized and a blood sample was obtained. The results showed a significant decrease in body weight if the mice were exposed to radiation. This was evaluated using the determination of testicular biomarkers in serum. Daily melatonin injections alone produced little change in testicular parameters. Exposure to gamma irradiation caused a significant decrease in sperm count, motility and vitality after 3 and 14 consecutive days and there was deformation of the head and tail, as well as a decrease in the concentration of LH, testosterone and melatonin with a significant increase in FSH levels. γ-radiation exposure to 4 Gy caused a decrease in androgen receptors and PCNA as well as a significant decrease in the antioxidant activity in testicular tissues. On the other hand, oral treatment of irradiated rats with melatonin for 3 and 14 consecutive days improved the levels of HNE-4, H2O2 and OHdG-8. in testicular tissue. The irradiated rats showed a significant increase in the serum levels of CRP and IL-6 with a significant decrease in the levels of IL-10 in the blood serum. The levels of Cyto-c, caspase-3 and p53 were significantly increased in the testes of rats. Oral treatment of rats irradiated with melatonin for 3 and 14 consecutive days reduced DNA damage and showed a significant decrease in tail length, percentage of tail DNA, and tail moment. In conclusion melatonin treatment reduced testicular damage caused by exposure to ionizing gamma rays and disrupted sperm parameters. |
