Abstract Bovine viral diarrhea (BVD) disease is a highly contagious and immunosuppressive worldwide cattle disease. It is caused by BVD virus (BVDV) that may cause high economic losses in beef and dairy cattle industries. The accurate BVD diagnosis is the cornerstone in any successful BVD control program. The specific natural anti-BVD antibodies based commercial diagnostic tools still have their limitations. Therefore, we aimed in this study to prepare BVD-nanomolecular imprinted polymers (BVD-nanoMIPs) based diagnostic tools (BVD-nanoMIPs labeled with ZnO nanocrystals and BVD-nanoMIPs based SPR nanobiosensor) to be applied instead of their commercial counterparts. The purity and crystallinity of bare nano ZnO preparations fabricated under different experimental conditions were examined using XRD. The XRD results showed the preparation of pure and crystalline nano ZnO. The results also revealed the enhancement of product crystallinity by the increase of NaOH content in the reaction mix. The shape and the size of all nano ZnO preparations were investigated using HR-TEM. The HR-TEM results also reflected the main adjusting role of NaOH in that regard. It was noticed that by increasing the NaOH content additional nanospheroid particles converted into a rod shape. The conversion of whole particles into long rod-shaped ones was found when using the maximum tested amount of NaOH content. The bare nano ZnO (fluorescent label) and the labeled BVD-nanoMIPs were tested by HR-TEM, Zetasizer, Zeta potential, and fluorospectroscopy. The results verified the success in the preparation of BVD-nanoMIPs labeled with fluorescent ZnO nanocrystals (shell and core like structure). Besides that, the SPR analysis confirmed the establishment of BVD-nanoMIPs based SPR biosensor with high affinity and specificity for BVD epitope template (BVDT). |
