Abstract 6. Summary There are three objectives of the present investigation: The first was isolation and identification of bacterial strains able to hydrolyze xylan under alkaline conditions. The second objective was production, partial purification and characterization of bacterial xylanase hydrolyzing xylan. The third objective was the application of xylanase enzyme for partial degradation of sugarcane bagasse. The salient features of the finding are outline below:- 1- Isolate alkaliphilic bacteria that hydrolyzed xylan, this step takes place by screening for the xylanolytic activity of bacteria isolated from water samples at Wadi El Natrun soda lakes in Behera and Abu Qir beach in Alexandria, in Egypt. About 8 isolates were recognized that hydrolysis birchwood xylan and three isolates marked CW1, CW2 and XA were found to be the most potent isolates. Only two isolates were selected for the study (CW1 and XA). 2- Both bacterial strains were identified morphologically, biochemically and 16S rRNA gene sequence analysis and identified as Bacillus halodurans. CW1and Bacillus halosaccharovorans. XA. 3- The phylogenetic analysis of the isolated was constructed in comparison with other related xylan hydrolyzers, neighbor- joining distance analysis of their 16S rRNA gene sequence. When comparing the 16S rRNA sequence of strain Bacillus halodurans. NS928R (Which belongs to the Bacillaceae family and genus Bacillus) result suggested this strain was genetically most closely |
