Abstract

Burullus lake includes two common wild plants; Phragmites australis(Cav.)and Typha domingensis(Pers)which are investigated in the biofuel production process. Both plants recorded high values from carbohydrates, cellulose and hemicellulose that could convert into bioethanol. Although, the bioethanol steps effectively produced a good mixture, lignin represents a problem in the hydrolysis of biomass.Virus-induced gene silencing (VIGS) technique was used to downregulate three lignin biosynthesis genes in Brachypodium distachyon by using barley stripe mosaic virus (BSMV) with Agrobacterium tumefaciens. The knockdown of targeted genes; BdPTAL, BdHCT1 and BdHCT2, besides GFP and BdPDS as a visual marker, produced a significant reduction in lignin that is
emphasized by staining, qRT-PCR and lignin concentration results.Key words: Burullus lake, Phragmites australis, Typha domingensis, bioethanol, VIGS,Brachypodium distachyon, lignin,BdPTAL, BdHCT1, BdHCT2, GFP and BdPDS
