Abstract The current thesis emphasized the role of cell culture implementations in pharmaceutical appraisal of novel chylomicron nanocarriers (CMs). The cell culture technique was successfully applied for assessment of parenteral and oral chylomicrons of two vital herbal actives “luteolin” and “berberine chloride” respectively. These novel chylomicrons were developed in an attempt to improve characteristics and anticipated performance of the loaded drugs. The work in this thesis was divided into two parts: <Part one: Development and Appraisal of Luteolin-Loaded Synthesized Chylomicrons for Enhanced Tumor Delivery In the current part, biocompatible synthesized chylomicrons were successfully developed as a promising approach to enhance luteolin injectable delivery potential. They were prepared using commercially available lipids and possess the advantage of preferentially up taken by liver hepatocytes following intravenous injection. Different assessment techniques for luteolin-incorporated chylomicrons were performed based on in vitro characterization, in vivo antitumor activity and cell culture techniques. Therefore, part one was sub-classified into two chapters, as follows: Chapter one Development and In Vitro Evaluation of Chylomicron vs. Phytochylomicron as Novel Nano-carriers For Improvement of Luteolin Delivery In the current chapter, Luteolin phospholipid complex (LPC) ‘phytosomes’ was successfully prepared by stoichiometric reaction between luteolin and Lipoid S100. Solubility studies of the formed phytosomes showed enhancement in both water and oil solubility. DSC, IR, XRD and release were further confirmed the formation of the complex of LPC. After that, these phytsomes were incorporated into optimized chylomicrons prepared by thin film hydration technique. Such integration lead to formation of novel double nanocarriers or phytochylomicrons. Full in vitro characterization of phytochylomicron were performed. Spherical vesicles of core and shell structure with 8% luteolin load were achieved under optimal conditions. |
