Abstract

1-Introduction: Cervical neoplasia is ranked second after breast cancer that cause female deaths globally.2- aim of the work:The aim of the study was produce Tip-1 gene knockout at the chromosomal level using CRISPR/Cas9 platform then investigate whether Tip-1gene knockout can affect the therapy resistance of human papilloma virus positive and negative cultured cervical carcinoma cells.3-Material and methods: Four human cervical carcinoma cell lines were used(c33a,c33aE6,caski and siHa) /CRISPER/CAS/RT-PCR/WESTERN BLOT/ FLOW CYTOMETRY 4-RESULT:Tip-1 gene knockout in monoclones of all cell line was associated with reduction in p53 gene expression level in comparison to the control. 5-Discussion: Reduced levels of Tip-1 directly correlated with reduced levels of p53 protein expression. 6-Summary and conclusion: Reduced levels of Tip-1 directly correlated with reduced levels of p53 protein expression. 7-Refrences:115 8-Abstract: Tip-1 directly correlated with reduced levels of p53 protein expression. This effect was seen in cells whether or not they expressed mutant (C33A) or wild type (CaSki) p53 or full or partial HPV genome (CaSki and C33AE6). These results suggest that Tip-1gene regulates p53 protein levels in HPV positive and negative cells by a novel and non-canonical means.
