Abstract Background: osteoporosis and cardiovascular disease (CVD) cause increased morbidity and mortality. Several epidemiological suggest a possible link in the pathogenesis both diseases. The aim of the study is to investigate bisphosphonates (Bps) Alendronate (AL) on dyslipidemia and the effect of the statins (Simvastatin) (Sim) on osteoporotic. Moreover, comparing the effects of using mono and combination therapy of Al and Sim on osteoporosis and dyslipidemia in ovariectomized (OVX) rats fed HFD. Methods: The rats were randomly divided to 5 groups: sham operated, OVX-HFD untreated, OVX -HFD treated with Al (3mg/kg/d), OVX-HFD treated with Sim (6mg/kg/d) and OVX-HFD treated with a combination of Al and Sim. Tested drugs were used for 4 wks started 8 wks after ovariectomy to test their potential antiosteoporotic efficacy and anti-dyslipidemic effect in OVX –HFD rats. Results: OVX-HFD untreated rats were associated with a significant increase in BW, serum total cholesterol, TG & LDL and significant decrease in serum HDL, significant increase in serum inflammatory markers [iNOS & TNFα] as well as significant increase serum leptin level. This was associated with histolopathological changes of isolated aortic ring indicating occurrence of dyslipidemia. Treatment of OVX-HFD with monotherapy Al or Sim induced significant treatment of osteoporosis and dyslipidemia in OVX-HFD. Comparing chronic treatment with monotherapy (Al or Sim) with the combination therapy (Al and Sim) in OVX-HFD came in favor to combination therapy which led to a significant decrease osteoporosis and dyslipidemic signs in OVX rats fed HFD. There was significant difference between combination and monotherapy in all tested parameters. Conclusion: Sim has a beneficial anti-osteoporotic effects and Al has beneficial antidyslipidemic effect. Moreover , the combination therapy of Al and Sim has more beneficial as an anti-osteoporotic and antidyslipidemic effect on osteoporotic dyslipidemic female albino rats. |
