Abstract This work was carried out to study the molecular and epidemiological aspect of zoonotic anisakid nematodes in fish marketed in Damietta Governorate. This was done in Animal Health Research Institute, Damietta branch. A total of 247 fish samples; 101 marine water fish, 101 freshwater fish and 45 salted fish were collected from Damietta. Marine fish included 20 Sardinellaaurita, 20 Scomberjaponicus, 22 Trachurusmediterraneusand 39 Mugilcephalus. Freshwater fish were 37 Oreochromisniloticus, 20 Cyprinuscarpio, 20 Clariasgariepinus and 24 Bagrusbajad. Salted fish were 25 Sardinellaaurita and 20 Mugilcephalus. Anisakid larvae were isolated and identified from marine water fish, freshwater fish and salted fish. Molecular study was conducted on a total of 35 larvae isolated from 4 different fish species by PCR and PCR-RFLP analyses. These confirm that the isolated larvae were belonged to anisakid family. The dendogram analysis and similarity index was done and it was found that the similarity between the isolates reached 100%, but it was 0% between some other. After performing some treatments on the viability of anisakid larvae, it was found that deep freezing at -20˚C for 15 hr, frying for 15 min, roasting at 250˚C for 35min, salting for 6 days after pre-immersion in saturated salt solution of NaCl for 4 days and smoking at 30˚C for 6 hr after salting for 48 hr inactivate anisakid larvae in Trachurusmediterraneus. Anisakid larvae were isolated from fish samples. This was confirmed by molecular techniques. The viability of larvae was assessed after various treatments. |
