Abstract
In this study, an ELISA Kit was developed for Detection of antibodies to Lumpy Skin Diseasevirus (LSD).Trial of isolation and characterization of LSD virus from skin nodules samples obtained from EL Gharbia Governorate in year 2009 in Egypt. Skin nodules were propagated in SPF fertile chicken eggs and in MDBK cell line. Titration of suspected LSDV was performed. characterization of suspected LSDV was carried out using TEM, IFAT and PCR. Screening of 254 serum samples from vaccinated cattle and 14 serum samples from infected cattle obtained from different governorates in Egypt using SNT .92 field serum samples of vaccinated cattlewhich tested by SNT were collected to test using prepared ELISA plate for detection and quantitative of antibodies against Lumpy Skin Disease Virus (LSDV). The evaluation was achieved to evaluate this assay by comparison with Serum Neutralization test (SNT). Sensitivity and specificity were measured for every test using ELISA test for detection of LSDV antibodies. Although the two assays have the same specificity, their sensitivity was 96.2% and 98.7% for SNT and ELISA respectively. In conclusion, the present study reports the development of an indirect ELISA kit for detection of anti LSDV antibodies and the success of this kit in the field application trial.
