Abstract

In this experimental study, the anaesthetic effect of lidocaine, thiopental and halothane administered on 11 day of gestation was studied. The DNA fragmentation test (comet assay) and the histopthological findings in the brain of neonates obtained from mothers administered the above mentioned drugs were also investigated.1- Anaesthetic effect.It was observed that mothers administered lidocaine 5mg/kg , the local anaesthetic effect persists for 40-50 minutes, the mothers administered thiopental50mg/kg was anaesthetized for 30-40 minutes, while mothers administered halothane depend on the maintenance period recommended for medical interference.2-DNA fragmentation test (comet assay)It was observed that the brain of neonates obtained from mothers administered lidocaine revealed an increase in the cell damage percent of about 34% on the other hand the brain of neonates obtained from mothers administered thiopental revealed an increase in cell damage percent of about 40% more over the brain of neonates obtained from mothers administered halothane induced an increase in cell damage percent about 44% 3- Histopathological findings The pathological alterations induced by using either lidocaine, thiopental or halothane, on the brain of neonates were more or less similar The brain of neonates subjected to the above mentioned drugs revealed perivascular and perivascular edema, necrotic changes of some neurons, meningitis, neurophagia,some blood vessels appear collapsed, congested blood vessels, area of lymphocytic aggregation (Encephalitis).Congested blood vessels with extravasation of red blood cells (focal haemorrahage) astrocytic proliferation, necrosis and demylination of axons was also observed.
