Abstract

In the present study seven vaccines were prepared
•Vaccine No.1 C. perfringens types B and D toxoid
•Vaccine No.2 C. pseudotuberculosis bacterin
•Vaccine No.3 C. pseudotuberculosis toxoid
•Vaccine No.4 C. pseudotuberculosis whole culture (bacterin + toxoid)
•Vaccine No.5 C. perfringens types
C. pseudotuberculosis bacterin
•Vaccine No.6 C. perfringens types
C. pseudotuberculosis toxoid
•Vaccine No.7 C. perfringens types
C. pseudotuberculosis whole culture.
with C. perfringens types B and D + C. pseudotubercqlosis whole culture (vaccine No.7).
On challenging the vaccinated and non vaccinated guinea pigs, all vaccinated animals resist the challenge with 100% protection. The unvaccinated guinea pigs showed abscesses in liver, lung and spleen.
The antibody titers of beta and epsilon in sera of vaccinated rabbits was high for vaccine No.5 and 6 while vaccine No ,.1 and 7 had the lowest titre for both types as determined by serum neutralization test and ELISA.
Testing of immune response in sheep vaccinated with previously mentioned vaccines except vaccine No.1 by using challenge ’test revealed that vaccinated groups showed lower mean number of abscesses 6.3, 2, 7.6, 7, 8 and 9 for the groups respectively in internal organs (lung, spleen and liver) compared to that of control group which was 53.3 in the same organs.
The result of double irnmunodiffusion test in sera of sheep vaccinated with the same six vaccines showed the presence of C. pseudotuberculosis antibodies in all sera of all vaccinated animals from the 2nd week and up to 181h weeks post vaccination and up to 22” weeks in group vaccinated with C pseudotuberculosis toxoid (vaccine No.3). Also this group showed the highest antibody titre by using microagglutination test. These antibodies began to decrease from the 10th week to reach zero level by the 22nd week in all groups except group vaccinated with vaccine No.3 in which the antibodies still present up to 26th week.
Serum neutralization test and ELISA for beta and epsilon antibodies revealed that sheep in vaccine No.5 showed highest titre of antibodies against beta and epsilon toxins while vaccine No.1 showed the lowest titre. The level of antibodies for all vaccines began to decline from 6th week to reach the lowest level of antibodies by 30th week for epsilon toxin and 54th week for beta toxin.
