Abstract

The objective of this study was to produce in vitro potato microtubers to be used as a basic material for seeds multiplication instead of importation with hard currency. Eight different cultivars of potato (Solanum tuberosum L. ) were preliminary examined in this study, which namely Spunta, Diamant, Armada, Arnova, Markis, Fontana , Prevento and Burn. This study was carried out to establish an automated low-cost production technique for producing microtubers of potato (Solanum tuberosum L.). Generally, Spunta, Diamant and Burn cultivars were surpassed the other cultivars in some record characters and selected for the main experiment. The results proved that the response of Spunta cultivar to the in vitro tuberization was more pronounced than those of Diamant and Burn cultivars. SDS-PAGE was used to detect some biochemical genetic markers related to microtubers production. SDS-proteins profiles indicated the presence of a band with molecular weight of 135 KDa in the three cultivars under the study which could be used as a biochemical genetic marker for in vitro tuberization in potato. The RAPD analysis indicated that each tested cultivar showed a complete genetic similarity between the two different sources of DNA which isolated -#102;-#114;-#111;-#109; tissue culture-derived microtubers and field growing tubers. br Key Words: Potato, tissue culture, microtubers, biochemical markers, SDS- PAGE ,molecular markers, Randomly amplified polymorphic DNA (RAPD-PCR)
