Abstract
Many identified diseases of the brain were proved to originate from disturbance in the binding interactions of dopamine to its CNS receptor sites. Decreased
binding might produce Parkinson’s disease, whereas elevated mode of binding could lead to mental disorders. In such cases, either dopamine therapy or dopaminergic blockade would be required. The main problem with dop— amine was the difficulty to cross blood-brain barriers, owing to its low liposolubility. The latter characteristic has always been taken into consideration when designing potential, centrally acting, dopaminergic agents.
