Abstract We can summarize this study as follows; Forty two ejaculated semen samples were collected from infertile couples enrolled for ICSI program due to male factor in Adam International Clinic. Routine semen analysis was done for each semen sample according to WHO guidelines. Aniline blue test for studying of the chromatin healthiness was done. Sperm DNA cytometry was done using the DNA content analysis. Sperm total genome was evaluated using gel electrophoresis for detection of damaged DNA bands. Pregnancy test was done two weeks after embryo transfer.This study results showed that; pregnancy outcome post ICSI may be associated with higher level of haploid sperm % and lower level of sub-haploid % in male factor infertility. Routine semen parameters (concentration, motility, morphology.aniline blue test, clinical parameters and fertilization rate cannot predict ICSI outcome. Embryo quality was better in pregnant group, with overall implantation rate of about 11% that negatively correlated with female age (r=-0.54), with early pregnancy loss rate of 36.8% (7/19) in the pregnant group. Haploid sperm% cutoff of 22.7% showed 94.7% sensitivity and 65.2% specificity for prediction of pregnancy in those patients. |
