Abstract

This work was done for studying of the antiprolifcrative effect of melatonin and Nigella sativa on the skin cancer model induced by the chemical carcinogen 7, 12 di-methyl henz anthracene (DMBA). The’ possible mode of actions for these drugs as anti-tumor drugs were also evaluated in this study.
Thirty mice were used in this study. The mice aged 50-60 days.
They were divided into four main groups as follows: - One group was the chemical carcinogenic (DMBA) group. The second group was the Control (not received any drugs). The third group was the treated group with melatonin and the fourth group was the treated group with Nigella sativa.
Biochemical results showed that DMnJ\ injection into the rats produced changes in thc levels or hoth apoptotic marker (caspase-3) and oxidative stress markers (lipid peroxide and Nitric oxide)
Daily administration of melatonin and Nigella as treatment after tumor growth produced signIfIcant decrease (in compare to the DMBA group) in tissuc Icvl’l or casp:lsc-.l indicating the role or appptosis as a mechanism or the anti-cancer ciTed or these drugs.
Oxidative stress plays an important role In explaining the carcinogenic elTed or the chemical carcinogenic and hence in correcting the carcinogenic state or the skin treated witll melatonin and Nigella saliva, where there were increase in the levels or hoth lipid peroxides and Nitric oxide in DM13J\ group in (ompare to the control group. Daily administration of the rats with melatonin and Nigella sativa were found to have antioxidative action through decreasing Nitric oxide levels in all treated groups both treated and prophylactic and decreasing the levels of lipid peroxides signifIcantly in case of lnelatonin and Nigella sativa.
